Position Summary

The Medical Technologist I in Molecular Pathology performs a variety of tests of a moderately or highly complex nature on patient and quality control samples following molecular pathology procedures. They use good laboratory practices to ensure that the identity of specimens is maintained throughout all stages of testing to provide the highest quality of results. They are expected to behave professionally, foster a culture of support and helping one another, and participate in continuous quality improvement.

Duties & Responsibilities

  • Perform highly and moderately complex tests using manual methods and/or various instruments in the Laboratory, on various specimen sources using established procedures under supervision. This includes participating in the development and validation/verification of new tests.
  • Hematology/Oncology molecular - manual qualitative nested PCR procedures for detection of fusion transcripts; semi-automated quantitative analysis for minimal residual disease in Chronic Myeloid Leukemia using Real Time PCR procedures; semi- automated allelic discrimination for V617F JAK2 mutation using Real Time PCR procedures; reverse transcription and nested PCR amplification followed by sanger sequencing to determine if mutations are present in BCR-ABL, causing resistance to tyrosine kinase inhibitor therapy; sanger sequencing or sanger sequencing and fragment analysis for detection of mutations in other oncogenic targets (MYD88, NPM1, FLT3).
  • Next-generation Sequencing (NGS) molecular- B-Cell IgH and MRD, T-Cell TRG, and Light Chain IgK gene rearrangement analysis to identify clonality.
  • Solid Tumor/Oncology molecular - manual qualitative nested PCR followed by sequencing to determine if mutations are present which may impact cancer treatment (KRAS, BRAF, EGFR); DNA extraction from tumor (colon cancer) and normal FFPE slides, multiplex PCR followed by capillary electrophoresis and fragment analysis to identify microsatellite instability (MSI) in the tumor.
  • Bone Marrow Engraftment Analysis molecular - semi-automated extraction of total, CD3, or CD33 DNA followed by PCR and electrophoresis through a capillary array. Short Tandem Repeat (STR) sequences are analyzed to discriminate between donor and recipient cells to determine the extent of donor engraftment in the bone marrow.
  • Genetics molecular- Mutation Detection by Sanger Sequencing for detection of inherited disorders (Factor V Leiden and Factor II); Taqman Single nucleotide polymorphism (SNP) genotyping for pharmacogenomic testing to determine patients who may be at risk for altered metabolism of drugs that are modified by cytochrome P450 2C19.
  • Infectious Disease molecular- Automated system for nucleic acid extraction, PCR amplification and detection to quantitate the viral load of blood-borne diseases (HIV-1, HCV, HBV) and transplant testing (CMV, EBV, BKV).
  • Perform routine molecular pathology procedures as needed:
  • Receive a variety of patient specimens arriving at the laboratory for testing and perform the steps necessary to prepare specimens for testing or referral to other laboratories as well as follow-up on inappropriate test orders.
  • Manual DNA/RNA extractions from whole blood, plasma, bone marrow, cerebral spinal fluid, and other body fluids as needed.
  • Automated Extractions: performs extraction of DNA and/or RNA extractions using an automated system and applies knowledge of extraction theory to troubleshoot system.
  • Preparation/Storage of reagents: prepares various reagents used for a variety of molecular techniques such as PCR master mix and stores appropriately; applies knowledge of various pieces of equipment used in the preparation of reagents or dilutions.
  • Use technical and scientific knowledge to review and analyze raw data for qualitative results and use expertise to analyze quantitative data to produce a report. Carefully review test result entries in the LIS before final verification. Consult with supervisor prior to the release of results when results are unusual or unanticipated.
  • Perform established quality assurance procedures (i.e. quality control testing, instrument

function checks and calibrations) and maintain appropriate records for these procedures. Complete Corrective Action Reports to thoroughly document any matter requiring follow-up or corrective action, including any attempted and failed runs.

  • Navigate the laboratory information system for all records, functions, and inquiries related to

testing of patient specimens.

  • Maintain a neat, orderly, and safe working environment in the laboratory while adhering to

all guidelines of OHSA''s Bloodborne Pathogens Control Act. Be knowledgeable of College of American Pathologists (CAP) checklists and inspections.

  • Interact with physicians, patients, staff, visitors and all customers via electronic messages, telephone conversations, and in person, in a pleasant and helpful manner.

Frequently check the "To Do List" in MediaLab and complete assignments in a timely manner.

  • Assist in instructing students and laboratory personnel in the principles, techniques, and analysis of

of diagnostic tests.

  • Perform other duties as assigned.
